Als neuen Ansatz zur Krebs-Früherkennung wird EDIM (Epitope Detection in Monocytes) verfolgt. Dabei werden die Tumormarker Apo10 und TKTL1 in CD14+/CD16+-Monozyten im Blut zytometrisch detektiert. Der Test kann mit hoher Sensitivität und Spezifität Karzinome erkennen, lässt jedoch keine Aussage zur Karzinomlokalisation zu. Eine Lösung dafür soll die Kombination mit dem spezifisch in Darmepithelzellen exprimierten Membranprotein GPA33 darstellen. In dieser Arbeit wurden monoklonale Antikörper gegen GPA33 hinsichtlich ihrer Eignung für den Einsatz im EDIM-Verfahren charakterisiert. Dafür wurden die Antikörper zytometrisch an GPA33 exprimierenden (HT-29) und nicht-exprimierenden (HEK293T) Zellen getestet. Dabei zeigte ein großer Teil der Antikörper eine sensitive und spezifische Bindung gegenüber GPA33. Weiterhin wurden Modellversuche mit GPA33 transfizierten monozytären Zellen (THP-1 und U937) durchgeführt, um zu überprüfen, ob sich das GPA33 Antigen auch in Monozyten detektieren lässt. Außerdem wurde versucht, bakteriell exprimiertes GPA33 nach Phagozytose transformierter E. coli Bakterien durch differenzierte U937-Zellen nachzuweisen. Die genutzten Antikörper waren auch nach Optimierungsversuchen nicht in der Lage, GPA33 in monozytären Zelllinien zu erkennen. Parallel wurden PE-konjugierte GPA33-Antikörper an menschlichen Blutproben anhand des EDIM-Protokolls getestet. Ebenfalls wurden Färbungen gegen die allgemeinen Tumormarker Apo10 und TKTL1 durchgeführt. In der statistischen Auswertung zeigte sich, dass fünf der sechs genutzten GPA33-Antikörper signifikant zwischen Proben der Gruppen „CRC“ und „kein CRC“ unterscheiden konnten. The study aims to assess whether AI systems demonstrate comparable or even superior levels of economic knowledge and to explore potential language-related differences in test performance. Descriptive statistics and item-level analysis are used to identify patterns and inconsistencies across languages when comparing AI test-takers and secondary students. Results show that AI models achieved near-perfect accuracy, while students performed significantly lower on average. The analysis also highlights differences in the length and explanatory depth of responses among the AI models. Findings contribute to current debates on assessment validity, AI use in economic education, and the future of multiple-choice testing in digitally augmented learning environments. Impairment is common in chronic pain, suggesting CPM may serve as a mechanistic indicator. However, the lack of protocol standardization and reference data prevents clinical use in individual patients. Methods We compared two CPM protocols with different conditioning stimulus intensities, test stimulus types, and interaction timing. We assessed CPM effect size, test-retest reliability and sensitivity to detect loss of descending inhibition. Results Conditioning with 0°C water led to stronger inhibition of pressure pain threshold (PPT) than conditioning with 7°C water (Cohen's d = 0.52), when tested immediately after conditioning. When tested during conditioning, effects of 7°C water immersion on heat pain sensitivity had similar magnitude (D = 0.53) and test-retest reliability (ICC = 0.77) as those on PPT (D = 0.54, ICC = 0.73). For all outcomes assessed, 95% confidence intervals (CI) of CPM effect included some facilitation instead of inhibition. The maximum degree of facilitation compatible with normal CPM (upper cutoff of CI) indicates potential sensitivity to detect individual abnormality. This was most favourable for PPT assessed after conditioning with 0°C water (decrease by more than 75 kPa or 14% of baseline PPT). Conclusions In conclusion, testing during conditioning stimulation yields medium to large effect sizes and good test-retest reliability. PPT testing immediately after ice water immersion has the narrowest 95% CI and hence offers the potential to generalize CPM assessments beyond group-level differences and compare inhibition among individuals in clinical practice. Significance Statement Indicating the main aspects where this work adds significantly to existing knowledge in the field, and if appropriate to clinical practice. Simultaneous CPM protocols exhibit large effect sizes but are confounded by divided attention.
